TXA2R mRNA and protein are present in AVCMs. A. 10x and 40x images of isolated AVCMs from male mice after 24 h in culture. Cells remain in high density; maintain membrane integrity and show clear striations associated with healthy cardiomyocytes following 24 h in culture. B. Real-time RT-PCR of RNA isolated from AVCMs showing the presence of TXA2R. Similar TXA2R gene expression was also observed with HL-1 cells. Inset shows TXA2R protein from isolated AVCMs detected by western blot. C. Representative data of 10 μM U46619-induced increases in intracellular Ca2+ in cardiomyocytes as measured by Fura-2 AM. This demonstrates that the TXA2R in mouse AVCMs is functional and behaves similarly to our previous reports in the rabbit [16].
